Nacho Libre is loosely based on the story of Fray Tormenta ("Friar Storm"), aka Rev. Sergio Gutierrez Benitez, a real-life Mexican Catholic priest who had a 23-year career as a masked luchador. He competed in order to support the orphanage he directed.

Calpuina is a night-watchman in a wharehouse, a perfect job for him. When some thieves break in and steal some crates, El Santo is after them because they are using the wharehouse for diamond smuggling. Capulina hinders their capture. Then he decides that he will put on a wrestler's mask and help El Santo to caputre the crooks.

At the dawn of the Second World War, Nazism was extending its grip over South America. Dr. Frankenstein and her assistant, Dr. Yanco, are going to bring one of their experiments to life in Santo Vs. Frankenstein's Daughter. She intends to use a monster named Ursus to do her evil bidding. Using a youth serum to retain their vitality, the doctors set their sights upon none other than El Santo. They need his super human blood to regenerate a stronger youth serum. They kidnap Santo's goddaughter, Norma and lure him into Dr. Frankenstein's lab where he is captured and enslaved. Will Santo make it out alive?

But when a psychopath wearing a wrestling mask begins butchering the teenage residents, their rehabilitation will become a no-holds-barred battle for survival. When Santo accompanies a troupe of archaeologists in the search for Aztec artifacts, they find the Mummy of an Aztec who leaves a warning to those who tamper with his resting place. The adventurers ignore the warning, and before long, the Mummy starts bumping them off one-by-one with his bow & arrow.

A group of international criminals steal a large emerald from a heavily guarded Colombian mine and they plan to sell it on the black market. Unfortunately, the evil Dr. Igor is also manipulating other criminals, as well as the ones who stole the emerald, for his own benefit. Santo, a Mexican wrestler/Interpol agent works with the Colombian authorities in an attempt to recover the emerald and to stop Dr. Igor and his subordinates, as well.
